Tryptamine
What is a tryptamine?
Tryptamine is an organic compound built around an indole ring (a bicyclic structure of a benzene ring fused to a pyrrole ring) connected to an amino group via a two-carbon chain. This core structure is called the tryptamine backbone.
What makes tryptamines important in psychedelic science is that serotonin itself is a tryptamine. This means that tryptamine-based psychedelics are structural relatives of serotonin — they fit into serotonin receptors because they literally look like serotonin to the receptor.
Tryptamine psychedelics
The most important tryptamine psychedelics for microdosing:

Psilocybin and psilocin
Psilocybin is a prodrug — it's converted in the body to psilocin, which is the actual psychoactive molecule. Psilocin is 4-hydroxy-DMT: literally DMT with a hydroxyl group at the 4-position. This minor chemical modification changes its receptor binding profile and duration of action compared to DMT.
LSD
While LSD is structurally more complex than simple tryptamines, it contains the tryptamine core within its larger molecular structure. This is why it acts on serotonin receptors and produces cross-tolerance with other tryptamine psychedelics.

Why tryptamine chemistry matters for microdosers
Cross-tolerance
Because tryptamine psychedelics share the same primary receptor target (5-HT2A), using one creates tolerance to all others. Taking psilocybin on Monday means LSD on Wednesday will be less effective.
Metabolism
Tryptamines are metabolized by monoamine oxidase (MAO) enzymes in the body. This is why:
- DMT is not orally active alone (MAO breaks it down in the gut)
- Ayahuasca combines DMT with MAO inhibitors to allow oral activity
- MAOIs can dangerously potentiate tryptamine psychedelics
Reagent testing
Different tryptamines produce characteristic color reactions with reagent test kits (especially Ehrlich reagent), allowing you to verify that your substance is actually a tryptamine before consuming it.
